Is the sub-contractor required to give a notice to the one who has only signed a promise to purchase a land without having signed a deed of sale?

The mere signature of a promise to purchase does not exempt the sub-contractor to give a notice of his construction contract to the beneficial owner. The tolerance of the land's owner for letting the construction begin is not either sufficient.
